Cytostatic agranulocytosis and concomitant infections are the often complications of intense 'e polychemotherapy in patients with acute leukosis. Human embryonic hemopoietic cells were used in a complex treatment of cytostatic agranulocytosis in 11 patients.
Cell suspensions, prepared each from one embryo (gestation terms from 6 to 12 weeks) cryopreserved with dimethylsulfoxid, were used. The material was tested on CFU-GM, CFU-GEMM, CD34, and underwent the viral and bacteriological control. The embryonic cells transplantations were performed intravenous drop-wise once or twice.
11 patients with cytostatic agranulocytosis were under observation. Hemopoietic cells were administered to 6 patients for the infection progress prophylaxis. At the trans- plantation moment 5 patients had the following complications: 1 - pneumonia, 1 - sepsis, 1 - angina, mucositis, 3 patients - fever without an educed infection focus.
During 3 days after the transplantation a positive dynamics was observed among all the patients with developed infectious complications: general state improved, intoxi- cation syndrome reduced, body temperature normalized (temperature of the patient with sepsis lowered to subfebrile). There were no infectious complications noted among the patients, who were administered human embryonic hemopoietic cells preventive.
Positive dynamics of peripheral blood parameters corresponded with positive dy- namics of blood clinical picture. At the moment of transplantation the leukocytes number was 0.96+0.17, granulocytes number 145.46+53.84. By the 5th day after transplantation, the leukocytes number was 3.44+2.05, granulocytes number - 203 5.69+ 1026.98. The sta- tistically significant rise of leukocytes number (p<0.05) was achieved by the second week after the transplantation (8-11 day).
Concomitant thrombocytopenia (50.7+11.37) was observed in 9 patients with agranulocytosis, 5 patients were observed for the hemorrhage syndrome signs such as dotty hemorrhage eruption, nasal bleedings. After the administration of hemopoietic cells positive dynamics of thrombocytes was observed. By the 7th day after transplantation thrombocytes number was 186.55+96.35, then more (p<0.05 by the 2 week).
Hemopoietic embryonic suspensions administration enabled to reconstitute hemo- poiesis of the patients, reduce the intoxication and carry on due chemotherapy according to the protocol.
